New Study Reveals Strategies to Shield Neurons from High-Fat Diet’s Adverse Impact on Multiple Sclerosis Progression

Dietary Choices and Multiple Sclerosis: New Insights

Recent findings published in the journal Glia have shed light on the relationship between diet and the progression of multiple sclerosis (MS). The study, led by Patrizia Casaccia, a prominent figure at the Advanced Science Research Center (CUNY ASRC) and an esteemed professor of Biology and Biochemistry, investigated the impact of certain dietary fats, particularly those found in palm oil, on neuronal health and the severity of MS symptoms.

Multiple sclerosis is characterized as an inflammatory autoimmune disorder that results in the damage of the myelin sheath, which insulates nerves across the body. While existing treatments primarily aim to modulate the immune system, the underlying factors that drive neurodegeneration in MS require further exploration. Earlier research from Casaccia’s lab had indicated a link between high-fat diets and increased MS severity. This recent study extends that inquiry by focusing on the toxicity associated with palm oil consumption.

Impact of Palm Oil on Neuronal Health

The research team employed the experimental autoimmune encephalomyelitis (EAE) model to simulate inflammation and demyelination. Their findings indicated that a diet high in palm oil resulted in a more severe disease progression in mice.

“We found that within neuronal cells, palm oil was metabolized into C16 ceramide, a toxic agent produced by the enzymes ceramide synthase 5 (CerS5) and ceramide synthase 6 (CerS6),” explained Casaccia. “This ceramide inflicts damage on the mitochondria, hindering the neurons’ energy supply necessary to combat brain inflammation. Hence, we examined whether disabling these enzymes could offer protective benefits.”

Upon genetically deleting these enzymes from neurons, the researchers observed a notable reduction in neurodegeneration in their MS model.

“Even when the mice were fed a palmitic acid-rich diet, the deletion of CerS5 and CerS6 still conferred protection,” remarked Damien Marechal, the study’s co-first author. “This highlights a specific metabolic mechanism through which certain dietary fats can exacerbate MS symptoms.”

Implications for Patients and Healthcare Professionals

The implications of these findings are significant for both patients with MS and healthcare providers managing the disease. The study underscores the importance of dietary choices in influencing the disease’s trajectory. It builds on previous research advocating for mindful eating habits as a component of MS symptom management and reveals potential molecular targets for mitigating diet-related symptom severity.

“Our study clarifies the molecular pathways that can be harnessed to protect neurons from the harmful effects associated with palm oil consumption,” stated Casaccia. “We aim to empower patients to make dietary choices that might positively influence their condition while exploring therapeutic strategies to counteract the impacts of CerS5 and CerS6 specifically within neurons.”

The research received support from the National Institute of Neurological Disorders and Stroke, part of the National Institutes of Health (NIH).
